Using a pooled CRISPR knockout screen targeting 195 genes with 799 sgRNAs, the study managed to identify regulators affecting NF-κB nuclear accumulation.
This spotlight poses as a great example of how Myllia’s technology combined with OPS allows for spatial target discovery. (4/5)

The first readouts from AVITI24 showcased:
✔️Cell compartments, with the nuclei highlighted in blue, the actin cytoskeleton in red and the cell membrane in green;
✔️sgRNA detection and assignment to cells;
✔️NF-κB localization through p65 antibody signal detection. (3/5)

NF-κB, a TF family involved in cellular signaling in response to cytokine stimulation, was an ideal pathway to study using this approach. Given its extensively studied nature, the involved genes were easily targeted, and its nuclear translocation easily visualized. (2/5)
