Not sure if any one noticed, but Lady Gaga's Super Bowl Half Time show (Salsa arrangement, 8 beat, 4/4 time signature) of "Die with a Smile" carries a much different vibe than the original 3/4.

I’d argue that in most cases it is inferred. Mutagenesis studies are quite clear for some states, and for the COs isotope exchange helps. But the terminal COs show strong coupling. I think 2DIR is pretty useful at being able to tackle much of this without needing to manipulate the system.

Overall, 2DIR is a fantastic technique that extends the incredible utility of IR spectroscopy methods applied to hydrogenases and other redox metalloenzymes. Not only can measurements be performed at ambient temperature, it is non-destructive, and high quality spectra are obtained in short time.

In this proof of concept study we apply 2DIR to the oxygen stable Hinact state of the [FeFe] hydrogenase from Desulfovibrio desulfuricans. 2DIR allows extraction of intra- and intermode anharmonicities. Combined with computations this provides exquisite insight into active site structure & dynamics.
