By combining two immune-boosting adjuvants alum and SMNP the vaccine lingers in lymph nodes for nearly a month, encouraging the body to produce a vast array of antibodies. This one-shot strategy could revolutionize how we fight not just HIV, but many infectious diseases. It mimics the natural infection process and opens the door to broadly neutralizing antibody responses, a holy grail in vaccine design. And best of all, it's built on components already known to medicine. Editor’s Note A great deal of research has aimed to improve immunogens for vaccines. However, improving immunogens alone will not necessarily result in a successful vaccine. Here, Rodrigues and Zhang et al. focused on the other half of the vaccine, the adjuvant. The authors aimed to understand why a combination adjuvant including a slow-release alum and a saponin-based nanoparticle, termed alum-pSer/SMNP, elicited considerably better immune responses to an HIV envelope trimer immunogen than either alum-pSer or SMNP alone. They found that alum-pSer/SMNP increased immunogen accumulation on follicular dendritic cells within draining nodes and activated a larger pool of B-cells with greater B-cell receptor diversity. These effects appeared to be complementary, resulting in an overall boosted humoral response to the HIV envelope trimer immunogen. Together, these data highlight the importance of adjuvants and demonstrate how tailoring adjuvants to vaccines could improve their efficacy. Abstract Vaccine adjuvants play important roles in shaping the humoral response to immunization. Here, we analyzed mechanisms of action of a clinically relevant combination adjuvant strategy, where phosphoserine (pSer)-tagged immunogens bound to aluminum hydroxide (alum) adjuvant, promoting prolonged antigen release to draining lymph nodes, are combined with a saponin nanoparticle adjuvant termed SMNP, which alters lymph flow and antigen entry into lymph nodes. When used with a stabilized HIV envelope trimer antigen in mice, this combined adjuvant approach promoted substantial enhancements in germinal center and antibody responses relative to either adjuvant alone. Using single-cell RNA and B-cell receptor sequencing, we found that the alum-pSer/SMNP combination augmented the clonal expansion and diversity of the germinal center B-cell repertoire, coincident with an increased proportion of S-phase germinal center B-cells and expression of positive selection markers. Moreover, we found that the combination adjuvant approach, but not alum-pSer delivery or SMNP alone, promoted accumulation of intact antigen on follicular dendritic cells, reflecting integrated effects of slow antigen delivery and altered lymph node uptake. Genetic ablation of Cr1/2 expression by follicular dendritic cells eliminated antigen accumulation and hampered the antigen-specific germinal center response, supporting antigen delivery to these cells as a key mechanism of the improved response elicited by this combination adjuvant. These results demonstrate how adjuvants with complementary mechanisms of action affecting vaccine biodistribution and kinetics can enhance humoral immunity. The research was funded by the National Institutes of Health; the Koch Institute Support (core) Grant from the National Cancer Institute; the Ragon Institute of MGH, MIT, and Harvard; and the Howard Hughes Medical Institute.
