On exposure to a cell-permeable, fast-binding JF dye ligand, neurons genetically expressing HaloTag become permanently fluorescently labeled, enabling precise visualization of targeted cells27,29 or, as recently shown, identification of newly inserted receptors or synaptic proteins30,31. We first performed a series of control experiments to determine key features and parameters of the CLOK labeling. In a first set of experiments, we examined if all genetically targeted neurons could be labeled with a JF dye in larval zebrafish using a transgenic (Tg) line co-expressing the HaloTag protein and a membrane-localized GFP. After incubation with a JF dye (here, JF525) at 5 days post fertilization (d.p.f.) (Methods), we evaluated the degree of colocalization of the dye and GFP fluorescence, finding 97.43 ± 1.8% of co-expressing neurons (Supplementary Fig. 1a; mean ± s.d., N = 3 fish, 16 planes), confirming robust, near-complete JF dye labeling. Delaying imaging after dye incubation in the same Tg line (incubation at 3 d.p.f. followed by imaging from 3 to 6 d.p.f.) increased the number of JF dye-negative, GFP-positive cells located at the periphery of the tectum (Supplementary Fig. 1b). This observation confirms that the HaloTag-JF dye complex remains stable, preventing JF dye from binding to newly expressed HaloTag...

Just got hit in my Gmail account - your date of birth is missing & we must comply with the law
Thank y’all, including for the info re: Proton. Thus far, there’s been no request for ID, but how much longer, I don’t know. Since user1 mentioned the 15gb gmail limit, I probably do have room to transfer all my years of gmails (of my 3 gmail accounts), but meanwhile, I had entered my birthdate in my junkmail account a few days ago, and thought I was out of the woods. But despite having entered my birthdate, just now, gmail asked me to enter the verification code sent to my recovery email (which is my “acquaintance” gmailbox). Then when i tried logging on to my “acquaintance” gmailbox, it asked me to enter my recovery email (which is my junkmail gmail address). They also asked me to enter my birthdate for my “acquaintance” gmailbox, after which i had to click CONFIRM that i’m age blah-blah… I finally was able to gain entry to both those gmail boxes. It just doesn’t stop. I assure you there is zero privacy in my 3 gmail accounts. It’s too late by now. The only thing which would ever recover privacy would frankly be a solar flare or something of the sort. P.S. I used to use both the vintage YahooMail and vintage Gmail. Then when i heard Verizon was taking over Yahoo, I panicked and asked around. At that time, lots of onliners advised me to transfer to Gmail, so i transferred my primary-Yahoomail and my junkmail-Yahoomail to separate Gmail accounts. I forget if i already had my acquaintance-Gmail account or created it at time of transfer. It was a major ordeal, but finally got done. Now i’m having DejaVu all over again, because Gmail has turned rogue for quite awhile already. Anyway, I guess that about wraps this up. Thanks again!!

Interesting sonnet 5.5 thing: In my personalization instructions. I have my birthdate so that claude can wish me happy birthday and remind me to update that number in there. Prior claudes never mentioned or thought about this not on that day. O5.5 always thinks about it, S5.5 always mentions it...

I have never bought or donated $$$ on FB. They don’t have my credit card info or real birthdate…

I also don’t integrate with other apps nor do I use fb login on other sites (and only sparingly with Google).

Not 100% fail safe but I am not going to make things easy either.
I won't even allow a Google Home or Amazon Echo in my home, and I've been burned by *password managers* and bad Chrome extensions; I sure as heck won't trust Meta's Muse to do anything involving personal information

No way to sign in to Bluerewards. Acc to them I got mt own email wrong(the one they spam me at), got my birthdate and postal code wrong, they won't send password reset emails(but claim to), won't amswer emails, and phone lines are closed at all times(even during posted "open" hours).
