Not sure exactly what you mean by inertia but (my color choice or its subsequent establishment) but I used red green because those were the dye we used (Cy3 and Cy5) were excited with red and green lasers.

I spend a lot of my free time thinking about lungfishes now. They don't have language. They live in mud puddles. They can grow to 4 feet long. If you trained an AI to act like a wild lungfish it would probably not enjoy pets and belly rubs, but aquarium lungfishes love pets and belly rubs.

You are right, it's not a BONCAT problem per se, it only becomes a problem for the FACS part of BONCAT-FACS. The dyes reacting with the peat was not a major problem, it was quenching and autofluorescence that made it tricky. We tested different azide-dyes and ended up using Cy5 as that worked best.

The Cy5 has 90% intensity falloff from center to edge of a well, just a 4mm radius away.
The collection optics have a vignette within the nominal working area, with a 75% drop in efficiency along that same radius.
And no proper aperturing for stray light control.
Unnecessarily compromised.
