It was so difficult that Vincent taught me how to CsCl band the low copy plasmid vector I was cloning into so I could make huge volumes of super pure concentrated vector because I had to optimize so many different parameters. Sisyphus except the rock is goddamn RV1A & the hill is cloning into pACYC.

I went to grad school back in Ye Olden Days at the dawn of the Qiagen maxiprep kit era. Prior to that, plasmids had to be separated from chromosomal DNA using CsCl density gradients. This involved loading DNA into tubes with CsCl and EtBr & spining in an ultracentrifuge overnight at very high speed.

Oooh I miss this. Yes, it's really simple anymore, especially compared to the old-skool CsCl method, and yet so amazing.
no matter how many times I’ve done it, isolating high-molecular weight DNA — so you see fibers by eye — always amazes me

you need salt, water, vinegar, & alcohol

salt & water occur in nature independently of life

vinegar & alcohol are products of metabolism (life)

To show my continuing love for VLPs: HPV-16 L1 VLPs, made in insect cells and purified on CsCl density gradient by ultracentrifugation, diafiltration, and ion exchange chromatography. And Eric van der Walt, now a hotshot at Roche in Cape Town, who made them in my lab in 2005.

I've talked about calcium oxalate (CaOx) in cacti, where they may constitute up to 85% of the dry weight of the plant. This allows the plant to manage excess calcium absorbed from the soil.

Pineapples & kiwifruit store CaOx in needle-like raphides, which may serve as defense against herbivory.

Nah, for real? CsCl was quicker, less effort, gave far higher yields and the quality of dna was infinitely better. Qiagen dna is like 💩, awful stuff. I miss CsCl. Same for their awful dna purification spin columns, everything is so dirty and performs so badly! Worst reagents in molecular biology!

Good question.
Points to the chemical form being CsCl - Caesium chloride, rather than being a ceramic form that is inherently more resistant to degradation..
Suggests it’s a orphaned radiotherapy source - the chloride form has been banned in industrial use for many years after contaminating accident
