#Cysteine
Pleased to share our recent work out today in @natmetabolism.nature.com. This study addresses a longstanding mystery - NRF2-driven cancers increase cysteine acquisition (via xCT) by ~5x - so where does all that cysteine go?

Why is #cysteine amino acid so toxic to cells?

www.rockefeller.edu/news/40469-c...

— intracellular cysteine stored as glutathione
— excess cysteine frees iron from ferritin
— excess iron in mitochondria damages iron-sulfur proteins
— mitochondria fail
— cell death

The Rockefeller University » Why high levels of the amino acid cysteine are toxic to cells
The findings explain why the cell prefers glutathione, and may lead to new therapies for certain types of neurodegeneration and cancers.

Check out our latest work out today in @natmetabolism.nature.com in which we describe a new metabotype (metabolic phenotype): excess cysteine stress. We discovered novel cysteine conjugates and show how pyruvate modulates free intracellular cysteine levels. Grateful for an awesome team!!

Thrilled to share our new paper @Chouchani Lab in Cell; what a great way to end this year! We establish a deep mapping of the zinc-binding human cysteine proteome (ZnCPT) and discover glutathione reductase as zinc-targetable cancer vulnerability. www.cell.com/cell/fulltex...

Ever wondered what the lonely histone H3C110 does? We found that it’s lost in many fungi. Adding it back boosts histone H3 copper reductase activity, rescues iron defects and modulates lifespan, suggesting an evolutionary tradeoff between these effects.
