Overall we conclude that Dreg1 is a Tcf1-dependent non-coding RNA critical for fine tuning the high level of Gata3 required for the optimal development of the ILC2 lineage. Enjoy!

Study of the chromatin landscape around the Dreg1 locus revealed that TCF1 regulates Dreg1 expression and that a syntenic GATA3 enhancer region on human chromosome 10 harbours novel noncoding RNAs with high expression in ILC2s.

Enter Adelynn Tang from @ajithvasanthan.bsky.social lab who found that loss of Dreg1 leads to a selective reduction of ILC2 in peripheral tissues. ILC2 are known to rely on high levels of Gata3 for their development and indeed we found a decrease in ILC2 progenitors in the bone marrow.

Dreg1 is a noncoding RNA we discovered a few years back that lies close to the classic Tce1 enhancer of Gata3. As Tce1 is important for T and NK cell development and Dreg1 mirrors Gata3 expression we were surprised when Dreg1 KO mice had normal T and NK cell development.
