Oxford Nanopore has updated their flowcell pricings: Minion largest discounts, some 30% cheaper than before. Flongle also cheaper. Promethion is now between $900 and $600 per flowcell.

The preprint of the first of the batch of parasitoid wasp genomes that we are generating is now out. Erythrina gall wasp is an invasive pest on the Hawaiian wiliwili tree, we were able to generate a HiFi genome using 1/10 of a flowcell, costing only ~$100 for the sequencing.

Nanopore sequencing 96 isolates on a promethion flowcell using the rapid barcoding kit= (900+330)/96 =13 $/isolate.
Cheaper if you get discounted flowcells from bulk deals.

Also, one bad flowcell and it eats your overhead. I don’t worry about bad flowcells with outsourcing

We’ve just discovered that NovaSeq clamps keeping the glass flowcell scratch FC surface releasing tiny splinters that are sucked by the pump. Given enough time, it will clog the instrument. #illumina #lablife 🧪

Realfreq, a framework for real-time base modification analysis for nanopore sequencing, is now published in Bioinformatics.
Written efficiently using C, a laptop can keep up with a
@nanoporetech.com MinION sequencer, and a desktop a PromethION 2 solo flowcell.

Expanding the genome evenly is hard because DNA is a polymer, so here we use the Buenrostro lab's favorite enzyme Tn5 to make fragments beforehand

We then do Illumina sequencing, but instead of on a flowcell, all enzymatics are performed ~inside~ the expanded nucleus

For sequencing curious folks: a lab is trying to use up their last NextSeq 550 flowcell, and they're willing to run it for anyone for cost of the flowcell or less (likely a lot less).

Anyone interested in 30~39Gb worth of 150bp reads, let me know! We have about a week to figure this out.

PacBio HiFi one flowcell, no HiC. Scaffolded onto the closely related K. laxiflora chromosome scale assembly available on JGI. Scaffolding was easy because most chromosomes are single contig already.

We get around 150Gbp of QCed seqences per flowcell, which would yield a coverage of 125x if the genomes are on average 3Mbp and evenly represented.

A) 5 sales per quarter is the current guidance going forward. Erik Holmlin was categorical when asked the question - the emphasis is on flowcell sales, management aims to be ultra prudent in its guidance at all times.
