Nice preprint! We still don’t know how insulin-responsive GLUT4 storage vesicles (GSVs) are made. @steveroyle.bsky.social shows a subset of intracellular nanovesicles sort and retain GLUT4, supplying components that build these unique vesicles.

NOT ONLY THAT.

Part of the reason their blood glucose is so high is they are insensitive to insulin, They don't have GLUT4, the glucose transporter that we have that responds to insulin by heading to the membrane to suck up sugar.

In their Opinion article, Makoto Kanzaki and Gwyn Gould re-exam the model of GLUT4 trafficking fat and muscle cells based on new studies suggesting that heterotypic fusion with endosomes might play a more important role than previously realised.

Our latest work showing that GLUT4 exofacial antibodies can measure translocation in humans and rodent skeletal muscle fibers and is impaired in TBC1D4 KO rats despite rescued GLUT4 expression. Congrats Kaspar and co-authors. DM me for open access version:

Grabbing hold of GSVs nuked the insulin-induced mobilisation, but capture of INVs only partially reduced it. So the INVs and GSVs are not synonymous. Instead it seems that INVs sequester GLUT4 and related cargos into GSVs to form the insulin-responsive pool. This is an endosomal sorting step.
