Automatic MS/MS Data Mining Strategy for Rapid Screening of Polyether Toxins Derived from Gambierdiscus Species #AC pubs.acs.org/doi/10.1021/...
Automatic MS/MS Data Mining Strategy for Rapid Screening of Polyether Toxins Derived from Gambierdiscus Species
Gambierdiscus is a primary producer of diverse polyether toxins that can biomagnify and transform within marine food webs, posing major risks to marine organisms and human health. Currently, many toxins derived from Gambierdiscus remain unidentified. Existing toxin analysis methodologies primarily rely on known toxins, limiting the representation of toxin diversity and complexity and potentially underestimating associated risks. Herein, we present a Toxin-Screening program for high-throughput screening of polyether compounds by analyzing MS2 fragmentation patterns of detected ions and identifying Pacific Ocean ciguatoxins (P-CTXs) and gambierones through specific ion recognition. Using the Toxin-Screening program, eight P-CTXs purified from fish and Gambierdiscus spp., alongside two commercial gambierones standards, were successfully extracted from 5027 MS2 spectra and annotated. This method was subsequently applied to profile polyether compounds in three Gambierdiscus caribaeus strains, revealing only ten polyether compounds shared among the strains, while strain-specific compounds dominated. All G. caribaeus strains were found to produce gambierone, with levels notably varying among the strains. Several polyether compounds containing one or two SO3 groups suggest a potential novel toxin family that warrants further investigation.

Toxic microalgae discovered in the Mediterranean could end up on your fish plate
A species of toxin-producing microalga has now been documented along mainland Spain’s Mediterranean coast, marking the first confirmed appearance of this organism around the Iberian Peninsula. Its arrival extends a food-safety concern, long associated with tropical seas, into waters that supply millions of people with fish. Coastal samples speak Coastal waters off Denia and Javea, along Spain’s southeastern Mediterranean shoreline, provided the first physical record of this Gambierdiscus organism outside tropical island waters. Gambierdiscus is a marine dinoflagellate that is a type of single-celled alga. By examining routine marine samples, Dr. Cesar Bordehore, an ecologist at the University of Alicante (UA), documented the presence of the toxin-producing genus directly in peninsular Mediterranean waters. Archived material from the same coastline showed no trace of the organism in earlier years. This means its presence is due to emergence within the past decade rather than being a long-standing feature. That narrow window frames the finding as recent and dynamic, which sets up the need to understand how far the organism may spread and what conditions allow it to persist. A toxin pathway Near-shore microbes such as Gambierdiscus can release ciguatoxins, nerve-acting poisons that enter into the tissues of grazing or filter-feeding fish. Small...

Theory is elevated levels of an algae known as Gambierdiscus, detected in the water column in areas with affected fish, as well as in the gut contents of some affected fish. The higher than normal water temperatures in the Keys last summer could have caused the spread of this algae undetected.
