Such natural IS110 diversity may expand our genome-editing toolbox. We validated an active 843-bp IS110 encoding a 201-aa recombinase, which is the most compact IS110 system shown to catalyze targeted insertion.

Excited to share our latest preprint from the lab! 🧬

We found that IS110 elements, typically considered small, can grow into large elements that carry cargo and disseminate antimicrobial resistance. We also experimentally validated the smallest and largest IS110s characterized to date.

Sun et al. show that IS110 transposons utilize two independent RNA-guided transposition pathways: a top-strand copy-out mechanism and a direct top-strand transfer to new targets. These findings redefine IS110 transposition and reconcile prior discrepancies, which provides insight into programmable, RNA-guided genome modification without double-strand DNA breaks.

We found that natural IS110 systems can span from ~800 bp to nearly 100 kb, which is far beyond the canonical ~1.5-kb model.

Two papers in Nature describe a new genome editing technique that enables the insertion, inversion and deletion of long DNA sequences at user-specified genome positions. A single-step approach could provide an easier method of genome editing.

IS110 elements can gradually grow by repeatedly gaining new DNA and duplicating parts, or even all, of the element, which allows them to accumulate cargo over time.

Expanded IS110s can also move across different bacterial lineages. We found about 30,000 cases of nearly identical IS110s in different species, especially among Klebsiella, Enterobacter, Escherichia, and Salmonella. Much of the DNA accumulated by IS110 appeared to come from plasmids.
