Another #Lifeact model system. the quail is still my favourite though… #cytoskeleton @melaniedwhite.bsky.social

How curious? This myeloid cell, even with its chromatins completely condensed, refuses to complete its mitosis. I thought this was my chance to image its division, but after awhile, it just moved away from me, even with its chromosomes still condensed! 🔵=MLC, 💜=H2B+LifeAct.

Happy #MicroscopyMonday.
I found this very pretty COS-7 cell expressing LifeAct-mCherry from my old data. Look at how dense its actin network is. Also, many of the moving dots are endosomal to seemingly push them through the cytoplasm, similar to actin comet tail, but just a lot shorter.

Cool!

HeLa cell with lifeAct, homebuilt 3D SIM.

(imaged 14 years ago in Gustafsson lab).

I'm willing - grudgingly - to accept that Lifeact is not a valid pangram in today's Spelling Bee, but what's wrong with felicitate?!

We confirmed this switch using a bunch of different techniques, from live imaging of transgenic choanos expressing an F-actin marker (LifeAct) to ultrastructural expansion microscopy, and the story was always the same: no actin cortex in flagellates, but a clear cortex after confinement.

So, there is a small (very small) 9-aa motif that is an evolutionary conserved anchor to actin.
It's named: short linear actin filament-binding motif (SFM), and it's *the same* found in LifeAct !

Kai wanted me to pass on his answer, it's LifeAct 😁

Thanks, Pablo. This is an early macrophage and its LifeAct 😁

Happy #FluorescenceFriday!
I captured this shot of a group of embryonic macrophages expressing LifeAct and H2B migrating inside tissue. These cells can use lamellipodia, filopodia, or blebs to migrate, and are frequently switching between these different modes or combining them. So fascinating.

Haha thank you so much. This is just LifeAct-RFP. The cell is COS7. I used Airyscan 880 to make this video.

Arittri's amazing finding that our old friend Lifeact can make actomyosin flow in the opposite direction, change chirality and flip left and right in the C.elegans worm.

#MicroscopyMonday Disco cell! One more RFP-LifeAct cell on top of nanoneedles. I believe these needles are actually a polymer cast of a cast of an Si array. I love the way the cell is exploring the topography with actin-rich protrusions that look like little octopus suckers. (5 min/frame)

#MicroscopyMonday Epithelial cells expressing RFP-LifeAct on an upside-down Si wafer. I used silicone grease to hold the wafer down on a 3D-printed support, and some spots of grease got on the wafer. These tiny bumps show up as bright rings of actin where the cells crawl over them! Fun accident.

To characterize these dynamics, we generated a new transgenic quail line expressing fluorescent reporters for the myosin regulatory light chain (MYL9) and F-actin (LifeAct). In both the epiblast and the PS, the reporters are strongly enriched at epithelial junctions. (2/n)

Witzig.

Then, we extended CALIPERS to human stem cells by expressing FUCCIplex and LifeAct in
Bruce Conklin's GCaMP6f-WTC11 hiPSCs reference line (parental line of the @alleninstitute.bsky.social Cell Collection).

FUCCIplex replaces GFP/RFP with iRFP & mTurquoise2, allowing multiplexed imaging of CC with standard fluorescence microscopes. See it in action tracking HaCaT cells across 40 hours (LifeAct in grey).

What is happening with these differentiating adipocyte-derived stem cells??? 🤯 Why is EGFP-LifeAct in/around the nucleus? Artifact?? Why would it disappear while others are stable for hours? #FluorescenceFriday #Actin
