Remarkably, Sandy found that the dI1-dI3s were specifically lost when netrin1 was overexpressed in chicken, while dorsal progenitors (dP) 1-dP3 expanded in the absence of netrin1 in mice. These cell fate changes correlated with the loss or gain of Bmp signaling respectively. 6/9

Sandeep Gupta took a bioinformatic approach using our dI stem cell model to assess the mechanism by which netrin1 regulates cell fate. He identified that netrin1 does not appear to act directly as a Bmp inhibitor but rather regulates mRNA processing. 8/9

Thus, the presence of netrin1 may stabilize the production of Bmp inhibitors, such as the inhibitory Smad, Smad7. Such inhibitors then suppress Bmp signaling and permitting the specification of the intermediate dI4–dI6 fates. In short, a really enjoyable Butler lab collaboration! 9/9

Work from many labs, including the Butler lab, has shown that Bmps act from the dorsal midline to specify the dorsal-most interneurons, dI1, dI2 and dI3. Moreover, there is a boundary of netrin1 expression immediately adjacent to the dorsal-most spinal cord. 5/9

However, Sandy rapidly hit a brick wall, because in the presence of high levels of netrin1 after in ovo electroporation, the commissural (dI1) axons disappeared. This is not a small phenotype; the axons completely disappear. 3/9

This paper grew out of an unexpected observation, by Sandy Alvarez, as part of her thesis work in the Butler lab. She was expressing tagged netrin1 in the chicken spinal cord, with the goal of live imaging the passage of netrin from progenitors -> pial surface -> axons. 2/9

This result was a mystery for months, until Sandy found a 2021 paper that suggested netrin1 could suppress Bmp signaling in vitro. She then set about assessing whether she had identified the same activity in the spinal cord in vivo. 4/9

Netrin1 blockade alleviates resistance to chemotherapy in pancreatic cancer
The study objectives and design are described in Methods. Between March 2023 and June 2024, a total of 43 patients from 9 centres were enrolled, including 3 in the safety lead-in phase of the study, where the dose of 14 mg kg−1 every 2 weeks for NP137 was confirmed. Demographics of the safety analysis population (n = 43) were consistent with general trends among patients with LAPC: median age was 65, 51% were male, patients had an Eastern Cooperative Oncology Group (ECOG) performance status of 0 (60%) or 1 (40%). Vascular encasement of aorta, mesenteric superior artery and celiac trunk was present in 5%, 58% and 51% of participants, respectively. Patients were considered at baseline as never resectable (54%) or potentially resectable (46%), with baseline CA 19.9 of more than 500 UI ml−1 in 42% of patients (Supplementary Table 1 and Supplementary Information). Of note, two patients were enrolled despite a metastatic disease. Safety is reported in all 43 participants who received at least one dose of treatment. All patients experienced at least one adverse event: 58% experienced at least one adverse event of grade 3 or higher, 47% experienced at least one NP137-related adverse event, including 12% who experienced...
