Our updated study is now published in Genes & Development! We set out to better understand how DAXX assembles H3.3 nucleosomes. This led us to identify separation-of-function mutants that allowed us to disentangle H3.3 deposition from ERV repression.

Finding the right gene to switch on is a needle-in-a-haystack problem, except the haystack is wrapped in nucleosomes.

Using minichromosomes, Wei et al. (2026) built a way to track single molecules of transcription factor GCN4 as it searched for its target gene.

The last part of my PhD work is on bioRxiv! Using Cryo-EM, optical tweezers, and in-cell footprinting, we showed that FoxO1 and HNF1α TFs engage and cooperate to bind nucleosomes in vitro and in native chromatin, modulating its architecture and stability. www.biorxiv.org/content/10.6...

Preprint: nucleosomes with active (H3K4me2) and repressive (H3K9me3) marks sit at distinct positions in plasma cfDNA. 50 of these sites read tumor fraction and neuroendocrine transformation from routine sequencing.

Different transcription systems shape chromatin behavior in distinct nuclear regions
Inside the cell nucleus, DNA is wrapped around histone proteins to form nucleosomes, which are further organized into chromatin. Chromatin has traditionally been divided into transcriptionally active ...

Mariani et al. in the Bulyk Lab at Brigham and Women's Hospital show that H3K27ac, H3K27me3 and H3K9me3 directly reshape how seven human TFs bind nucleosomes, shifting FOXA1 and PAX7 monomer-dimer preferences, with H3K27ac blocking internal sites to favor end-binding.

Rotational settings quantize nucleosome movement by chromatin regulators
Chromatin regulators are thought to override intrinsic DNA sequence preferences. La et al. show regulators move nucleosomes in quantized 10 bp steps to take advantage of the shallow energy potential c...

PARP1 directly disassembles nucleosomes to regulate DNA repair
Verma, Zhu, et al. show that PARP1 promotes NAD+-dependent eviction of H2A-H2B dimers, generating oriented hexasomes with increased DNA accessibility. The H2A C-terminal tail supports this remodeling activity, linking PARP1-dependent nucleosome disassembly to cellular responses to DNA damage.

🧬PARP1 isn't just a DNA damage sensor. It actively evicts histone proteins H2A-H2B dimers to transform dense nucleosomes into open hexasomes to regulate DNA repair. This motor-free remodeling links basic chromatin biology to targeted cancer therapies

mChIP-seq for multiplex epigenomic profiling uncovers cancer-specific histone features in cellular and circulating nucleosomes
Sun et al. develop mChIP-seq, an epigenomic profiling technology that enables multiplexed and multifactorial ChIP for both cellular and cell-free samples. They demonstrate the technology by profiling cancer-specific features of histone marks in cancer cells and in plasma cell-free nucleosomes from breast cancer patients.
