There’s a lot of really good research going on right now in orexin replacement, for both N1 and N2!

In the meantime though, we’ve just gotta keep managing how we can.

Researchers in South Korea found that SARS-CoV-2 selectively disrupts the brain’s orexin system, leading to lasting cortical neuron damage.

Boosting orexin levels reversed this effect, revealing a possible root of Long COVID fatigue and brain fog.

for some reason down a rabbit hole of mid-80s-mid 2000s discoveries (Leptin, Orexin, Growth Hormone production, cold receptor, p21 KO mouse, human iPSCs, etc) and realizing that they were all accepted less than two week after submission. Orexin in 4 days! Those were the days....

A cat will sleep for around 16 hours a day, given the chance. Labradors lack the appetite regulator orexin. I think I must be a good 40% cat, 40% labrador.

Patients with narcolepsy type 1 have hypersomnolence caused by a loss of orexin-producing neurons. Research findings from the TAK-861-2001 trial on oveporexton, a highly selective oral orexin receptor 2 agonist, in these patients are summarized in a new Quick Take video. nej.md/4mdFZUS

Excited to share new research from my lab in final published form! By deleting hypocretin (orexin) receptors from CRF neurons, we uncovered neuropeptide mechanisms linking alcohol drinking and negative emotional hyperarousal through BNST neuronal excitability.

4) Something similar has been reported in type I narcolepsy, a rare autoimmune disease that targets orexin neurons. Due to a lack of orexin, these patients fall asleep uncontrollably during the day and suffer from sudden muscle weakness.

I wouldn't say they're entirely unrelated. There's increasing evidence ADHD involves orexin dysregulation to some degree, and narcolepsy is caused by orexin deficiency.

45% of those with #LongCOVID suffer from a sleep disorder.

It's so prevalent it's dubbed 'coronasomnia'.

Overall, CSF orexin-A levels are crucial for diagnosing and understanding various sleep disorders and neurological conditions.
