Some stretches of our genome encode mysterious peptide sequences of very short amino acid length that lack matching versions in other organisms, so are omitted from proteome databases. In a new @nature.com paper, the TransCODE Consortium dubs these ‘peptideins’ & provides a framework to study them.🧪

Could these #peptideins also be important? We think Yes. And some might be on the path to our understanding them as full proteins.

We used extensive CRISPR-based analyses to show how these can display important cellular phenotypes in a number of contexts.

A new study reveals 1,700+ protein-like molecules hiding in the human genome — expanding what we thought we knew about the proteome.

Big picture: we find that lots of protein entities can be robustly confirmed in the human proteome. Some look like "real" proteins and we now annotate them as such. Most look like a mixed picture, with experimental evidence but not fully meeting "protein" criteria. We call these, #peptideins.

What these dark proteins--now renamed peptideins--are doing inside the body is not easy to answer. Some act like actual proteins, but others have much murkier roles, if any. But if one is closely associated with a specific cancer, for instance, we can use it as a target regardless of its role.
