🚨 New pre-print! If you are interested in new roles for SARM1, check this out. The lab did cool work showing macrophages need Sarm1 to efficiently respond to immunoogical stimuli and PNS injury. Cell specific sarm1 KO mice give surprising results- check it out!

(1) NMNAT2 is a repressor of SARM1 NADase in axons. A few people have NMNAT2 LoF mutations. Many more have low expression.

SARM1 inhibition protects neurons in FUS-ALS, Parkinson’s & excitotoxicity models. Will be exciting to see trials for these inhibitors in an expanding spectrum of neurodegeneration. Amazing therapeutic potential from inhibiting a protein originally found in a Drosophila mutant screen.

I’m Michael Coleman, working in Cambridge, UK on how nerve injury, and some gene mutations, toxins & viruses kill axons through the NMNAT2-SARM1 pathway, how to block this & which human diseases and specific patients it would be most useful to block it in.
