Therbusch was born in the Kingdom of Prussia in 1721. She completed more than eighty-five verified portraits. Her works often feature detailed depictions and strong lighting effects.

Anna Dorothea Therbusch wurde von Kurfürst Karl Theodor von der Pfalz 1764 zur Hofmalerin in Mannheim ernannt. Sie malte außerdem die Supraporten der Spiegelgalerie des Neuen Schlosses in Stuttgart (zerstört) und wurde 1762 Ehrenmitglied der Académie des Arts in Stuttgart,
