A few years back we discovered a dual hybrid protein modification composed of ADP-ribose dinucleotide and ubiquitin (ADPr-Ub). Now we reveal that ADPr-Ub can be further ubiquitinated by the E3 ubiquitin ligase RNF114!

Excited to share our latest work, now out in @lsajournal.org. we show that the E3 ligase HOIL-1 can ubiquitinate many different saccharides and describe a chemoenzymatic platform to generate ubiquitinated sugars as tools for studying non-proteinaceous ubiquitination.
The RBR E3 ligase HOIL-1 ubiquitinates Ser/Thr-residues and various disaccharides in vitro and can be used to generate ubiquitinated sugars as tools to study non-proteinaceous ubiquitination

'The paper focuses on one of these, ubiquitinated glycogen, and that phrase will probably be new to most people (it sure was for me). I had no idea that carbohydrate polymers like that could be ubiquitinated at all, much less that it’s apparently a major process for glycogen handling and disposal.'

Glycogen is ubiquitinated in vivo! And there is a clever method to detect it.

It had been a privilege seeing this story come together by my colleagues in the @komanderlab.bsky.social. This will no doubt have important implications for metabolism.

A RAD18 SAP domain PIP motif enables PCNA mono-ubiquitination and USP1-BRCA1 synthetic lethality
The proliferating cell nuclear antigen (PCNA) sliding clamp is a central component of eukaryotic DNA synthesis. In response to DNA damage, PCNA is mono-ubiquitinated by the RAD6 (E2)-RAD18 (E3) complex. However, the structural basis by which RAD18 engages PCNA to direct mono-ubiquitination has remained poorly defined. Mono-ubiquitinated PCNA can subsequently be extended with K48-linked poly-ubiquitin chains that target PCNA for degradation. Ubiquitin-specific protease 1 (USP1) reverses both mono- and poly-ubiquitination of PCNA; accordingly, USP1 inhibition promotes accumulation of mono-ubiquitinated PCNA at replication forks and reduces total PCNA levels, leading to replication defects that are synthetically lethal with BRCA1 deficiency. Here, we combine computational and structural approaches to identify and characterize a SAP domain PCNA-interacting peptide (PIP) motif within RAD18. We demonstrate that this interaction is required for DNA damage-induced PCNA mono-ubiquitination and for PCNA turnover following USP1 loss. Disruption of the RAD18-PCNA interface suppresses ssDNA gap accumulation and reduces USP1 inhibitor sensitivity in BRCA1-deficient cells. Furthermore, cells adapted to prolonged USP1 inhibition exhibit reduced RAD18 levels, suggesting that deregulation of PCNA mono-ubiquitination represents a biologically relevant resistance mechanism. A new paper from Dr. Edwin Miranda, who investigated the effects of suppressing oxidative stress by inhibiting Keap1. Results were somewhat unexpected, but highlighted the importance of nuanced roles that ROS play in muscle function. More papers to come on this related topic!
