Thanks to the Dr. Ruth Report for alerting us to another paper on WASF3. Further evidence of the potential importance of this molecule as a cause of fatigue and brain fog in people with ME/CFS and LC.

New Paul Hwang paper! 👀

“This study shows that stress-inducible Wiskott–Aldrich syndrome protein family member 3 (WASF3), previously linked to mitochondrial dysfunction in skeletal muscle of patients with ME/CFS, also suppresses oxidative metabolism in the brain.”

NIH researchers found that elevated WASF3 disrupts mitochondrial energy production in mouse brains.

Removing WASF3 improved metabolism and protected cognition under stress, revealing a potential mechanism for brain fog in ME/CFS and Long COVID.

"...Studie zeigt, dass das stressinduzierbare Protein WASF3 (Wiskott-Aldrich-Syndrom-Protein-Familienmitglied 3) – das bereits mit mitochondrialen Funktionsstörungen in der Skelettmuskulatur von ME/CFS-Patienten in Verbindung gebracht wurde – auch den oxidativen Stoffwechsel im Gehirn unterdrückt."

good to confirm existing science but pretty sure I've been reading about WASF3 for SEVEN YEARS now

they need to start working on dedicated drugs for it

because it's obvious now repurposing existing drugs isn't going to do it

new drugs will take a decade to get to market

do not have decade left

So the infection triggers the endoplasmic reticulum spiral and dysregulates WASF3?

WASF3 damages the electron transport chain, reducing ATP production and increasing ROS.

Increased ROS levels in #LongCOVID patients may loop back and damage the ER, creating a vicious circle:

I know! I was pretty siked about the WASF3 discovery, but it’s just been yet another dropped ball.

So NIH continues down this path of devoting resources to studying T cell exhaustion, WASF3 in subset of intramural cohort but also effort preference framed by behavioral science. I am not sure what to make of this. Also how has the funding status changed since this presentation was made?

"She wrote to a scientist about her fatigue. It inspired a breakthrough."

This is the background story to the recent study which reported excessive levels of the WASF3 protein in ME/CFS.
