a , Geometry of the metaphase spindle (spindle length, L and width of the metaphase plate, W) with 19 chromosomes (deformed spheres) and the forces exerted on the individual microtubule bundle (orange arrows), F left, F right and F chr , shown in side view (left) and front view (right). The force on the chromosome, F chr , arises from the repulsive force of the nearest neighbors, F. The positions of the spheres are calculated by the model, while their deformations are shown schematically; see the example of two spheres in contact with the indicated decrease in distance between the centers of the spheres, h , and the radius of each sphere, R. b , Width of the metaphase plate versus the number of chromosomes and the adjustment. The value of the lattice parameter is a 0 = 0.0025. The colored dots represent the experimentally measured average widths of the metaphase plates for: nearly haploid HAP1 cells (Supplementary Table 2), diploid metaphase I (MI) and haploid metaphase II (MII) human oocytes (Supplementary Table 2), and diploid and tetraploid RPE1 cells, as well as hypotetraploid RPT3 cells (Fig. 2 and Expanded Data Figure 2). c, Metaphase plate width versus chromosome number and model results. The sphere radius and chromosome number are varied simultaneously, while the total spindle volume is kept constant and consistent with parameter values N = 37 and R = 1.25 μm. The colored dots represent the experimentally measured average widths of the metaphase plates for the respective species, whose genome size is 5-8 Gbp (Supplementary Table 1). Inserts (1) and (2) show schematic coils with 7 and 37 chromosomes respectively (b and c). d, Phase diagram of the width of the metaphase plate as a function of the Young modulus and the chromosome radius. The yellow and red lines show values of R and E yielding W = 15 μm and W = 8.58 μm respectively, while the white lines correspond to the coil width values shown in the graph. e, Interchromosomal thrust versus chromosome number,… a , Examples of two solutions of the model without (left) and with (right) the external force exerted on the chromosomes, indicated by red arrows. The compression coefficient is k = 100 pN μm −1 . The solutions are shown in the front view (top left) and two side views (top right, height of the metaphase plate, W y , and bottom left, width of the metaphase plate, W z ). b , Interchromosomal thrust at the outer chromosomes, at y = 0 and z = 0, versus compression coefficient, k . c , Model prediction for the width and height of the metaphase plate versus compression coefficient, for two different sphere radii (shown in the legend). d , Angles between the outer microtubule bundles at the pole in two directions, α y and α z (see diagrams, k = 100 pN μm −1 ) versus compression coefficient. e , Spindles before, during, and after compression in live-photographed U2OS cells expressing CENP-A (color-coded for depth) and α-tubulin (microtubules (MTs), gray) with labeled DNA (cyan) in control cells (above) and MUG cells (below). f , The spindle compression setup. g , Spindle height (gray) and spindle width (cyan) before, during, and after compression for control cells (above) and mitotic cells with unreplicated genome (MUG) (below). h , The angle between pole and outer kinetochore fiber for three time points related to compression of U2OS control cells. Thick lines: mean; shaded areas: standard error; thin lines: individual cells. Number of cells: 12 (g , above and h ); 11 (g , below). The number of cells is equal to the number of independent experiments. In g and h : Student's two-sided t-tests. In figures a - d, the parameters are the same as those in figure 3, unless otherwise stated. Scale bar: 5 µm. a , Live-photographed RPE1 cells expressing CENP-A–GFP and centrin–GFP (color-coded for depth with circled centrioles) and stained with SPY650 DNA (cyan) in untreated control (top), treated with hypertonic solution (middle), and treated with hypotonic solution (bottom). The times indicate the start of imaging. Yellow arrows represent the coil width and white arrows the coil length. b , Coil length (top) and coil width (bottom) in the control (Ctrl) and at the indicated time points after addition of hypertonic (left) and hypotonic (right) solution. The colored points represent individual cells; black lines show the mean, with light gray and dark gray areas marking the 95% confidence intervals for the mean and the standard deviation, respectively. Number of cells: 16, 27, and 20 per treatment from 5, 10, and 7 independent experiments. Percentages indicate the differences between the average values of the groups. A diagram of spindle and acute cellular responses to hypertonic (bottom left) and hypotonic (bottom right) solutions. Control spindles are shown in lighter colors and treated spindles in darker colors (legend, left). Orange arrows indicate cell contraction in hypertonic solution relative to the control. Gray dots represent extracellular solute. c , Model prediction of spindle width, WS = W − 2 R , for different spindle lengths. d , Mean ± standard error for spindle width versus spindle length for protein perturbation experiments. Data from ref. 72. The number of cells per condition is between 7 and 39. Black line, linear adjustment ( R² = 0.58, P = 1.4 × 10⁻²⁵ ); gray area, 95% confidence interval. e , The spindle width is determined by the chromosomes and scales with their size. The spindle length is determined by microtubules (MTs) and microtubule-associated proteins (MAPs) and scales with cell size. In c, the parameters are the same as those in Fig. 3, unless otherwise stated. Inh., inhibited; KO, knockout; oe, overexpression; si, small interfering RNA. In … When the length of the DNA increases n times, the volume of a typical chromosome also increases n times, while the length and width of the metaphase plate increase only n 1/3 times. A substantial change in the size of the genome therefore requires only a small variation in the size of the spindle. The mechanism of spindle adaptation to varying genome sizes is based on the widening of the spindle due to interchromosomal push forces (arrows).

I have created new horrible women and I'm making it y'all's problem
illustration of Renata, a tall, serious business butch with a black and white undercut and a prosthetic arm and Nica, a shorter, fat butch with tan clothes. They're in a dimly lit room in front of a table with glow-y tech equipment. Nika is reaching between Renata's legs from behind, teasing: "Do you want to keep explaining the tech we both know I know how to use or can we get to the fun part already?" Renata looks annoyed, but is backing into Nica, calling her impossible. Nica just hums in agreement, asking: "So..?"
