Going nuclear: Improved antisense oligonucleotide activity through conjugation with a nuclear importer
Antisense oligonucleotides (ASOs) are a promising class of therapeutics designed to modulate gene expression. Both key mechanisms of action for ASOs operate in the nucleus: splice-switching ASOs modif...

'Unlike typical gene therapy, which alters a person’s genes to treat a genetic disease, antisense oligonucleotide [RNA] therapy, uses short strands of genetic material to target RNA produced by the gene and reduce how much protein is made.'

Looking for good news. Advances in RNA science & med.
First for RNA therapy: man with rare motor-neuron disease improves after treatment
Gene-targeting treatments could be used to treat other people with rare forms of amyotrophic lateral sclerosis (ALS).

strategy to reactivate TRA2β’s poison exon using antisense oligonucleotides (ASOs). These synthetic RNA fragments are designed to bind precisely to the TRA2β RNA, forcing the poison exon back into the sequence. When introduced into cancer cells, ASOs restored the natural SELF-DESTRUCT mechanism.

Had to look up an old paper to get the doi. It still completely boggles my mind that TWO Nobel prizes have been given for small RNAs and this work wasn't recognized by either. Abstract is literally like "yeah, we found these tiny antisense RNAs and think they're probably causing gene silencing"
