We first assessed aging in both oxidative soleus (SOL) and glycolytic extensor digitorum longus (EDL) muscles and found that CL levels declined similarly with age across muscle types (Fig. 1a and Extended Data Fig. 1a). We then compared the expression of CL synthesis and remodeling enzymes. Despite trends for Pgs1 and Taz to be decreased (Extended Data Fig. 1b), the gene most significantly reduced in SOL and EDL encoded the CL synthase, Crls1 (Fig. 1b and Extended Data Fig. 1c). We found a similar pattern in aged human muscle, with total CL levels declining in aged (53–69 years) versus young (22–35 years) participants, alongside significant age-dependent alterations in individual CL species composition (Fig. 1c and Extended Data Fig. 1d). Transcriptional analyses similarly revealed a pronounced decrease in genes linked to CL synthesis (Fig. 1d and Extended Data Fig. 1e), pointing to a conserved age-associated regulation of CL biology.
