and this signals into the PE cell and turns off nanog and sox2. (see my Thread 1 for more info on nanog and sox2). It also activates genes sox17 and gata6. Mainly keep in mind that primitive endoderm markers are oct4 and sox17. (5/n)

To do so, we generated transgenic mESC lines in which different types of synthetic enhancers (i.e. “typical” enhancer, CTCF-associated enhancer, 4x enhancer cluster) were built upon the same “core” enhancer and inserted at the same distance from a developmental gene (i.e. 100 Kb from Gata6)

Low GATA6 appears to switch colorectal cancer cells into a more primitive, metastasis-ready state. In mouse models, deleting GATA6 increased liver metastases with little effect on primary tumor growth. doi.org/hb8f7w

Meantime, EPI cells send FGF ligand to other cells but they don’t have the receptor for it themselves. So there is no way to suppress nanog & sox2. Nanog is free to actually suppress gata6 (so *really* preventing a PE fate)! Mainly keep in mind from this that epiblast markers are oct4 & sox2. (6/n)
