Sustainable oxime production via the electrosynthesis of hydroxylamine in a free state - Nature Synthesis
A sustainable two-step strategy for oxime synthesis in a neutral KHCO3 medium is reported. Free-state NH2OH is initially synthesized via nitrite electroreduction over a Ketjen-black-supported iron phthalocyanine catalyst, followed by a chemical aldehyde or ketone oximation using the electrochemically synthesized NH2OH.

Enabling Unfavorable Hydroamination Reactions Using a Chemoselective N–O Bond Reduction
Despite major advances, intramolecular alkene hydroamination reactions often face limitations. Herein, a redox-enabled process featuring oxidation of an amine to a hydroxylamine, a concerted hydroamination step, followed by catalytic reduction of N-oxide is shown to be broadly applicable. Catalyst screening and optimization showed that a K2OsO2(OH)4–pinacol complex rapidly and chemoselectively reduces the N-oxide cycloadduct in the presence of hydroxylamine and dimethyl sulfoxide. This selectivity was exploited to drive the equilibria toward complex products.

Do people generally make up fresh hydroxylamine or use it from a stock? If it’s the latter good chance it’s degraded. Also, NHS has a short half-life. Bottom line, is quenching TMT even necessary? Who has looked?

I'm particularly interested in how these properties in cytochromes P460 are affected by the unusual heme-protein crosslinking. This makes them green, distorts the heme and allows them to oxidise the iron-bound NH2OH to N2O, a greenhouse gas. This is basically what makes them #P460 instead of #P450!

Nitrene generation and transfer for unnatural biosynthesis in living cells
Having observed products of apparent nitrene transfer, we sought to incorporate both the generation and reaction of this nitrene into an artificial biosynthetic pathway in vivo. To do so, we envisioned that the enzymatic oxidation of PABA to generate the corresponding hydroxylamine N-OH-PABA and acylation of this hydroxylamine could be conducted in vivo before transfer of the nitrene catalysed by a cytochrome P450 (Fig. 3a). We tested a series of arylamine oxidases (AOs, including BezJ from Streptomyces sp. Rl18 (BezJRI18) and S. niveus (BezJniv) and AurF from Streptomyces thioluteus32,33,34,35) (Fig. 3b) that would oxidize the amino group in PABA in vivo (Fig. 3c). BezJniv was chosen as an alternative in case the low in vitro activity for BezGRI1817 was also seen for BezJRI18. AurF was included because it catalyses the oxidation of PABA to p-nitrobenzoic acid via the hydroxylamine, which we could trap by acylation33; as such, AurF functioned as a positive control for BezJ activity. To catalyse the acylation of the resulting hydroxylamine, we used BezGniv.
