Replication-uncoupled MET1/DNMT1 activity shapes inheritance of DNA methylation
Using live-cell sensors and duplex sequencing, Edera et al. show that Arabidopsis MET1 maintains CG methylation beyond DNA replication, methylates accessible nucleosomal DNA, and acts partly independently of hemimethylated sites. This nucleosome-constrained de novo activity helps restore lost methylation and shapes its short- and long-term variation.
