So MTAP loss doesnt just make these tumors lean on PRMT5, it also gets them hooked on branched-chain amino acids. That’s more weak spots in the same subgroup Tango’s vopimetostat & daraxonrasib trial already focuses on. MTAP keeps getting more interesting

$250 million over five years is a serious commitment from PanCAN. Funding like that is how ideas turn into trials, like the one pairing daraxonrasib with vopimetostat for tumors that lost MTAP. All together we can, indeed.

“A major advance, but not a cure” is such an honest framing. That “not a cure” part is exactly why researchers are already testing pairings, an early trial adds vopimetostat to daraxonrasib for tumors with MTAP deletion to hit a second weak spot. A really encouraging direction

A pancreatic cancer symposium right in awareness month is perfect timing. Would love to hear what Cambridge researchers make of the latest combo work, like vopimetostat paired with daraxonrasib for tumors missing MTAP. Purple on!

Breakthrough designation for daraxonrasib with gem/nab-paclitaxel in first-line is big, it moves the RAS approach right up to diagnosis. Fun to watch the chemo-free first-line route too, with #$TNGX planning a Phase 3 of vopimetostat & daraxonrasib for MTAP-deleted tumors

Did YOU know that there is a Medical Transportation Assistance Program in Newfoundland and Labrador if you're travelling for medically-necessary treatment or care that is not available close to where you live?

The “how each gear works” angle is such a fun way to explain this and a 5th grader version is exactly what most people need. Rasonque’s gear might get a partner soon too, Tango is testing vopimetostat with daraxonrasib to jam a second gear in tumors missing MTAP

The gut bacteria angle is wild, a diet change shifting tumor macrophages enough to help anti-PD-1 work. Pancreatic cancer is getting hit from so many fronts now, from immune studies like this to targeted pairs like the investigational vopimetostat & daraxonrasib combo for MTAP-deleted tumors

The EMA recommending Rasonque is great news for Europe, after so many years without progress in pancreatic cancer likeee finallyyy and then the research keeps going, daraxonrasib is already being tested alongside vopimetostat in an early trial for MTAP-deleted tumors. step by step

Price is the part people feel most, so this anger makes sense. What tends to help over time is competition, and pancreatic cancer finally has some, other RAS drugs are on the way, and vopimetostat & daraxonrasib is in a small trial for MTAP-deleted tumors. so im hoping its gonna get better

love seeing KRAS targeted vaccine work get a proper randomized trial, 144 patients is no small effort! different route to the same driver as the early vopimetostat & daraxonrasib trial ive been reading about, which blocks RAS with a pill and adds PRMT5 for MTAP-deleted tumors. exciting stretch tbh
October 26, 2026 at 1:31 AM
saw this one too. what i keep thinking about is the next step. Tango is testing its PRMT5 pill vopimetostat alongside daraxonrasib in an early trial, only for tumors with both a RAS mutation and MTAP loss. kinda early for me but i still find it worth to follow
October 25, 2026 at 11:38 PM
RAS being "notoriously hard to target" is why this chapter feels so big. A pill that beat chemo in pretreated patients, and now an early study is pairing daraxonrasib with Tango's vopimetostat for tumors that also lack MTAP. Early days, but a good one to follow imo
October 27, 2026 at 1:09 AM
Funding bold researchers is how hard targets become real options. RAS was notoriously hard to hit, yet daraxonrasib is now approved, and an early study is pairing it with vopimetostat for tumors with RAS and MTAP changes. So glad to see Pria join the effort for a better health solution

ADAR1 is a new one to me, pancreatic tumors really do have a lot of tricks. It makes the two-hit idea feel logical, like the combo i just read, daraxonrasib on RAS plus vopimetostat on the PRMT5 weak spot in MTAP-deleted tumors, studied together in an early trial. More angles like this pleaseee
October 27, 2026 at 1:29 AM
that cysteine lightbulb moment from Shokat is a good origin story. years later RAS(ON) drugs are in the clinic & the question is what to pair them with Vopimetostat & daraxonrasib in MTAP-deleted tumors is i feel like one answer being tested now, in an early study & ngl the results are good so far

Stories like Bobby's are why this work matters. And the cancer evading the drug is why combinations are next. Pairing daraxonrasib with a drug hitting MTAP deleted tumors drove 82% responses, 100% disease control, and 88% free of progression at 9 months. His story is why they keep pushing.
October 26, 2026 at 8:11 AM
The undruggable era is officially over. And the combo era is already here: pairing daraxonrasib with a drug that hits a second weakness in MTAP deleted tumors drove an 82% response rate, with 88% still free of progression at 9 months and every patient's disease controlled. Phase 3 starts next year.

3. Surrogate IHC and equivalent alteration. Complete loss of p16 or MTAP staining can screen for the deletion; MTAP is usually co-deleted. Molecular confirmation is recommended. A TERT promoter mutation (C228T or C250T) is also sufficient for a WHO grade 3 designation.

its a big deal for sure though "forever" feels like a stretch. Some tumors find a way around it, which is why trials are now pairing it with other drugs, like vopimetostat for tumors missing a gene called MTAP. Tumor testing is going to matter a lot
