Prior work by the @martinowk.bsky.social lab and others have shown that CST (along with other neuropeptides) are regulated by BDNF-TrkB signaling. Indeed, Aaron found that disrupting TrkB signaling in CST neurons is sufficient to reproduce the sex-specific effect of accelerated kindling rate.

Recent evidence suggest that both drugs also bind and activate an entirely different receptor, TrkB, the receptor for BDNF and a key driver of neuroplasticity.

When these drugs activate TrkB, they enhance the ability for neurons to form new connections, a very good thing for a depressed brain. 🧪

Case study 2: a fragment model built for 3′ chemistry, cross-fitted across donors, reveals an 8-donor shift in NTRK2 (TrkB) terminal-isoform usage: full-length share 20% in astrocytes/NSCs → 78% in mature neurons. Matches known TrkB.T1 biology. No reprocessing.
