Q for #chromatin BSky - We're doing Cut&Run on cells where we want to halt transcription at various short time points after induction, with a cold PBS wash. This cold shock seems to hinder Ab uptake during, so our libraries look crappy. Any tips? Alt. methods to stop transcription?

That is very interesting!
I wonder if mRNA decay factors play a role in this process.
For instance, Xrn1 was identified early on in yeast to function in strand exchange & homologous recombination.
Pat1 interacts physically with topoisomerase 2 in yeast.

I’m thrilled to share a collaborative story from @Mendell_lab and Jan Erzberger labs, led by postdoc extraordinaire Xiaoqiang Zhu. We showed that in addition to their canonical decoding function, tRNAs play a key role in regulating mRNA stability during translation!
