🤝 SWI/SNF was thought to prep chromatin then step aside. #Yeast cryo-EM reveals it stays, forming a 7-megadalton complex with the pre-initiation machinery, Mediator & the promoter nucleosome to actively stabilize transcription initiation

We compared spindles across 25 eukaryotic species, from yeasts to plants and animals: Metaphase plate width scales with genome size to the 1/3 power. A vast range of genomes, handled by a small change in spindles. Surprisingly, chromatin amount counts, not chromosome number.

We put this idea to the test. From polyploid human cells and patient-derived cancer organoids to physical compression of cells, experiments showed that changing chromatin content or crowding changes spindle geometry as predicted.

Beyond thrilled that the first paper from our lab is out today in @nature.com! We show that SWI/SNF loss results in gain-of-function for CTCF and cohesin, reshaping the 3D genome, and an unexpected switch between chromatin remodeling complexes.

Full thread below.

Loss of function of canonical BAF induces gain of CTCF and cohesin binding that alters 3D genome architecture and reprograms transcription.

This work started with a very simple and intriguing question: how can microtubules apply sufficient force in the nucleus to promote chromatin mobility, in particular DNA double stand breaks? 🤔 🧵
