ECO-CAIRS represents evolution in use of allogenic ring segments
The management of keratoconus has evolved substantially over 3 decades. While corneal transplantation was historically the only surgical option for advanced disease, intracorneal ring segments (ICRS), first implanted by Walton Nosé in 1993 and later popularized by Joseph Colin, offered a less invasive way to improve corneal symmetry and reduce irregular astigmatism. Conventional ICRS are made of synthetic PMMA, and despite their efficacy, they carry substantial risks, including extrusion, corneal melt, neovascularization and stress-induced stromal necrosis, particularly when placed

Macrophages transfer functional proteins to their surface during live-cell uptake
While analyzing a published single-cell (sc)RNA-seq dataset from the 4T1 mouse mammary carcinoma model20,21, we noticed that transcripts for a canonical epithelial-cell-restricted protein, epithelial cell adhesion molecule (EpCAM), were detectable in a subset of TAMs (Extended Data Fig. 1). This was consistent with a report observing human epithelial markers in RNA-seq of macrophages from an allogenic A549 xenograft mouse model; the authors suggest that this RNA is indicative of phagocytosis of human cancer cells22. By flow cytometry, a subset of macrophages in 4T1 tumors stained positive for surface EpCAM (Fig. 1a and Supplementary Fig. 1a). EpCAM positivity was only observed on macrophages isolated from tumors and not from bone-marrow-derived macrophages (BMDMs) (Fig. 1a and Supplementary Fig. 1b), indicating that a tumor-microenvironment-specific mechanism was responsible for tumor-restricted EpCAM being present on TAMs. Curious as to the origins of EpCAM transcript and protein, we engineered 4T1 cancer cells to express intracellular GFP and evaluated CD45+CD11b+Ly6G− TAMs that had or had not ingested 4T1-derived GFP in engrafted tumors (Fig. 1b). We observed increased anti-EpCAM surface staining of GFP+ TAMs compared to GFP− TAMs (Fig. 1b and Supplementary Fig. 2), suggestive of an association between cell uptake and anti-EpCAM staining. We reasoned that additional...

Posterior Segment Risk Factors for Penetrating Keratoplasty Failure
Corneal transplantation is the most common allogenic transplant performed worldwide.1 Although the use of Descemet’s membrane endothelial keratoplasty (DMEK) and Descemet’s stripping automated endothelial keratoplasty (DSAEK) has increased for the treatment of corneal endothelial disorders, full-thickness replacement via penetrating keratoplasty (PK) remains a common method of transplantation for pathology involving the stroma or the entire cornea.2 Commonly reported indications for PK include keratoconus, postinfectious corneal scarring or thinning, and pseudophakic bullous keratopathy.
