I've been working to visualize the biogeography of the aphid's intracellular symbiont within whole insects. The pink bacteria live in the host's specialized bacteriocytes and embryos. Really excited about where this work is going! #FluorescenceFriday

They're so interdependent with the aphid that they've lost the ability to make a functional cell membrane without help & have genomes 1/6th the size of their free-living cousins.

.. And no evidence of relaxed selection on protein-coding genes shared between Buchnera and Serratia, suggesting that the presence of Serratia does not exacerbate genome erosion in Buchnera even on redundant metabolic genes! ...

Next, @nancy-moran.bsky.social on the interaction between Buchnera and its aphid host. SyeA, an effector predicted to interact with Rho1 GTPase, is secreted during the host colonization. Knocking it down disrupts the aphid's embryo development

Yba3 (sorry, SyeA) was one of those enigmatic Buchnera proteins: (almost) always there, but who knows what it did.... Now we know! SyeA is "secreted and homologous to secreted effectors of bacterial pathogens AND is essential for Buchnera transmission"!

Really cool work!

finally came across a native WA #Orobanchaceae! #Buchnera ramosissima or linearis! Very exciting! Not much info on these plants, but growing in close proximity to grass I’m guessing that must be a preference! Other species tho are known to parasitise trees, but these species can live independently

They have one job: taking plant sap the aphid ingests & converting non-essential amino acids into 9 amino acids the aphid needs to survive (but cannot synthesize).

There's evidence genes necessary to do this conversion were captured from a FUNGUS this BACTERIA also infected!
