T cell cross-priming by PD-L1+ type 1 conventional dendritic cells mediates the antitumor efficacy of a PD-L1×4-1BB bispecific antibody
Bispecific PD-L1×4-1BB antibodies under clinical development enhance antitumor T cell responses. Luri-Rey et al. find that the efficacy of PD-L1×4-1BB antibodies relies on PD-L1-expressing type 1 conventional dendritic cells, which integrate antigen cross-presentation, IL-12, and 4-1BB costimulation to promote efficacious T cell-mediated antitumor immunity.

CTLA-4 is a key immune checkpoint molecule blocking CD80/CD86-mediated costimulation, and particularly important for Treg function.

Common CTLA4 genetic variants are associated with susceptibility to multiple autoimmune diseases, but how they affect human Treg biology has remained unclear.

NKG2D overexpression potentiates CAR-T cells through costimulation by tumor NKG2D ligands
Li et al. show that activated CAR-T cells upregulate NKG2D ligands (NKG2DLs), rendering them susceptible to NK cells. Overexpressing NKG2D shields endogenous ligands and harnesses tumor NKG2DLs for costimulation, thereby enhancing CAR-T cell persistence, tumor infiltration, and antitumor efficacy across multiple models.

New JEM study @mengzelyu.bsky.social & Sonnenberg (doi.org/10.1084/jem....) reports a counterintuitive result: blocking B7 costimulation promotes, rather than suppresses, the microbiota-specific RORγt⁺ Foxp3⁺ Treg cells. From @cytokinesociety.bsky.social: signals.cytokinesociety.org/2026/10/06/l...
Special Article of Interest by Mengze Lyu in JEM - Biweekly newsletter of The Cytokine Society
Blocking B7 costimulation was expected to dampen microbiota-specific RORγt⁺ Treg cells. Instead, it expanded them. Read Mengze’s own account of the surprise.

This is a consequential omission: ‘The DMARD type most often left out was T-cell costimulation modulators, of which there is only one, Orencia . . . . Among plans available for this year, it's covered in only 21.2% of Medicare Advantage plans and by a shocking 0.3% of standalone Part D policies.’
