With @amyhbuck we wrote this piece on extracellular RNAs that intends not to give answers but to propose questions that can expand the field in new directions.

Hope you like it! We really enjoyed a lot brainstorming it.

To identify regulators of neuronal exRNA export, we devised a genome-wide CRISPR screening strategy in which secreted sgRNAs link each cell’s genotype to its extracellular RNA phenotype.

The screen revealed thousands of potential exRNA regulators.

So how might Ψ drive exRNA export?

One possibility is that RNA-binding proteins recognize Ψ-marked RNAs and channel them into secretory pathways.

Surprisingly, we found that MYL6 (myosin light chain 6) selectively binds Ψ-containing exRNAs and is required for their extracellular transport.

We know very little about the function of extracellular RNAs (#exRNAs), especially in the context of brain biology.

And even at a more fundamental level, the mechanistic rules governing mammalian exRNA trafficking remain largely obscure.

Small RNA Fragments Hold Major Promise in Advancing Cancer Treatment
A groundbreaking review from Universiti Putra Malaysia and its regional partners is challenging the boundaries of cancer diagnosis and treatment by highlighting the transformative potential of extracellular RNA (exRNA) in oncology. These minuscule RNA fragments, many of which traverse the body encapsulated within exosomes, are capturing the imagination of researchers due to their dual utility: they are not only biomarkers signaling the presence and progression of malignancies but also vehicles capable of delivering precise therapeutic payloads directly to cancer cells.
