Thanks for replying: I know this paper, Rick Cummings referred me to one on pig kidney fucosidase. Still searching for a really very first paper on this one. As Sigma, NEB and Agilent/Prozyme no longer have it, perhaps we’ll have to express the bovine one (which sequence did you use for modeling?)

Bovine kidney fucosidase, the general purpose enzyme (except for core 1,3-Fuc) is no longer available from Sigma! But does anyone have a citation for its first isolation/characterisation. I have searched in vain!

Activity of lysosomal exoglycosidases in blood serum and urine of patients with tick-borne and other lymphocytic meningitis. bioRxivpreprint
Activity of lysosomal exoglycosidases in blood serum and urine of patients with tick-borne and other lymphocytic meningitis.
Tick-borne encephalitis (TBE), and other (non-tick) lymphocytic meningitis diseases (LMD) are characterized by the presence of inflammatory processes differently located and with different intensity. Inflammatory processes in TBE and LMD may influence the activity of some catabolic enzymes including lysosomal exoglycosidases: N-acetyl-{beta}-D-hexosaminidase (HEX), its isoenzymes A (HEX A) and B (HEX B) as well as {beta}-galactosidase (GAL), -mannosidase (MAN), - fucosidase (FUC) and {beta}- glucuronidase (GLU). The aim of the study was to investigate the activity of lysosomal exoglycosidases in the serum and urine of patients with TBE and LMD. Material and methods: Examined group included 41 patients: a/ 25 patients with TBE, b/ 16 patients with LMD. Control group included 25 patients in which Borrelia burgdoferi and inflammatory state in central nervous system were excluded. Lysosomal exoglycosidases activity were assayed in the serum and urine by the colorimetric method, with the p-nitrophenyl derivative of appropriate sugar as substrate. Results: Concentration of activity (pKat/mL) of HEX, HEX A, HEX B and GLU increases significantly in serum of patients with TBE, and LMD as well as GAL, MAN and FUC serum activity in LMD patients in comparison to serum of patients from the control group (p

I know that NEB chose another fucosidase doi.org/10.1038/s415... precisely because BKF is incompatible with AQC (rapifluor-like) tags (Chris Taron worked with us on the JPR paper 😎)

Aminoquinoline Fluorescent Labels Obstruct Efficient Removal of N-Glycan Core α(1–6) Fucose by Bovine Kidney α-l-Fucosidase (BKF)
Here we report evidence that new aminoquinoline N-glycan fluorescent labels interfere with the release of core α(1–6) fucose from N-glycans by bovine kidney α-l-fucosidase (BKF). BKF is a commonly emp...

Thought it would be simple to find but even in 1994 they were purchasing it from Sigma.
It looks like human kidney fucosidase was the first to be characterised, maybe that’s the best place to start?

α-L-Fucosidase Isoenzymes: New Glioma Prognostic Markers
In a groundbreaking study published in BMC Cancer, researchers have illuminated the pivotal role of α-L-fucosidase isoenzymes, FUCA1 and FUCA2, as prognostic markers in gliomas, a group of aggressive brain tumors. Gliomas, particularly lethal in their most malignant form known as glioblastomas, have long posed a challenge for effective diagnosis and treatment. This comprehensive investigation melds vast genomic data with experimental validation, revealing a new dimension in understanding glioma biology and patient prognosis.

🔹 Fucosidase
Estaban muy ligadas al contenido en azúcares y antioxidantes. ¡Casi se podían “leer” los genes en la química!
