Single-cell analyses of global cohorts outline determinants of immune aging and link the ratio of GZMK+ to GZMB+ Tem cells to health trajectories
Terekhova et al. assemble a single-cell PBMC atlas of 8 datasets, 2,609 individuals, and over 12 million cells to dissect variable and conserved features of human immune aging. CD8+ Tem GZMK+ to GZMB+ ratio distinguishes healthy from disease-associated trajectories and predicts mortality risk. This work provides a clinically relevant marker for monitoring human aging.

Researchers mapped immune cells from 2,609 adults. The GZMK-to-GZMB T-cell balance tracked health, while protein signatures linked to later disease risk. Observational evidence, not a clinical aging test.

The authors analyzed T and related cells from the ME scRNA-seq atlas, consisting of 28 patients and 30 sedentary controls at baseline and after symptom provocation.
Differences; decreased expression of effector genes, GZMB, FcGR3A, KLRF1. This may decrease pathogen control.

New research suggests a simple immune-cell ratio, the balance between GZMB+ and GZMK+ memory T cells, could become a new biomarker for biological aging. A history of CMV infection is a major risk factor.

Human CD4+ & CD8+ T regulatory cells play distinct roles in regulating autoreactive immune responses. Investigated FOXP3, GZMB, CRISPR. #ImmunologyInsights PMID:39806065, Nat Immunol 2025, @NatImmunol @OTSociety https://doi.org/10.1038/s41590-024-02062-x #Medsky #Pharmsky #RNA 🧪
Differential roles of human CD4+ and CD8+ regulatory T cells in controlling self-reactive immune responses | Nature Immunology
Here we analyzed the relative contributions of CD4+ regulatory T cells expressing Forkhead box protein P3 (FOXP3) and CD8+ regulatory T cells expressing killer cell immunoglobulin-like receptors to the control of autoreactive T and B lymphocytes in human tonsil-derived immune organoids. FOXP3 and GZMB respectively encode proteins FOXP3 and granzyme B, which are critical to the suppressive functions of CD4+ and CD8+ regulatory T cells. Using CRISPR–Cas9 gene editing, we were able to achieve a reduction of ~90–95% in the expression of these genes. FOXP3 knockout in tonsil T cells led to production of antibodies against a variety of autoantigens and increased the affinity of influenza-specific antibodies. By contrast, GZMB knockout resulted in an increase in follicular helper T cells, consistent with the ablation of CD8+ regulatory T cells observed in mouse models, and a marked expansion of autoreactive CD8+ and CD4+ T cells. These findings highlight the distinct yet complementary roles o

In IL-12.aFOLR2.CAR–treated tumors, cancer cells are closely juxtaposed with GZMB⁺ CD8⁺ T cells, consistent with antigen-dependent cytotoxic engagement. But that's not the only way the cancer cells die! To find out more, see the paper for full data, figures and the amazing co-authors!

"Although not observed in mouse ILC2s, we found that human ILC2s secrete granzyme B (GZMB) and directly lyse tumor cells by inducing pyroptosis and/or apoptosis, which is governed by a DNAM-1−CD112/CD155 interaction that inactivates the negative regulator FOXO1."

PERCISTRA revealed distinct regulatory mechanisms of BET and HDAC inhibitors. BET inhibitors suppress AP-1 activity and promote a TCF7-driven naive T-cell program, while HDAC inhibitors activate RUNX3/TBX21-associated enhancers, increasing GZMB and IFNG expression and promoting cytotoxic programs.
