Terekhova et al. assemble a single-cell PBMC atlas of 8 datasets, 2,609 individuals, and over 12 million cells to dissect variable and conserved features of human immune aging. CD8+ Tem GZMK+ to GZMB+ ratio distinguishes healthy from disease-associated trajectories and predicts mortality risk. This work provides a clinically relevant marker for monitoring human aging.

A study of 2,600 individuals found that the ratio of GZMK+ to GZMB+ T cells predicts health outcomes. Genetic and environmental factors drive differences in immune aging between people of the same age.

Researchers mapped immune cells from 2,609 adults. The GZMK-to-GZMB T-cell balance tracked health, while protein signatures linked to later disease risk. Observational evidence, not a clinical aging test.

New research suggests a simple immune-cell ratio, the balance between GZMB+ and GZMK+ memory T cells, could become a new biomarker for biological aging. A history of CMV infection is a major risk factor.

Fantastic preprint showing how circulating EBV-specific CD8+ T cells in MS adopt a GZMK+ effector–memory state that is primed to migrate to the CNS & may drive neuroinflammation through non-cytolytic interactions with B cells & myeloid cells. 🧵👇 1/

Functionally, activated CD8⁺ T cells promoted atypical B cell and plasmablast formation, while GZMK-enriched effector-memory cells induced inflammatory IL-6 and IL-8 responses in myeloid cells.

We found that CNS accumulation was not random. T cell clones carrying a GZMK⁺ effector-memory program already in peripheral blood were strongly biased toward the CSF. Strikingly, EBV-specific T cells were particularly enriched for this tissue-oriented state.

Incredible to see this elegant antigen-resolved approach converge on the same B cell-dependent GZMK-effector CNS-trafficking T cell state we triangulated via a reverse translational approach using B cell depletion & trafficking blockade in our recent @brain1878.bsky.social paper. 2/

Cool recent paper on immune triads.
As other papers have suggested, Cxcl13 and Gzmk are uber important for beneficial anti tumor interactions between T cells and APC. Also, triads are apparently common even without therapy!

However, in contrast to GZMK+ cells, the ISG population was transcriptionally inert. i.e they poorly responded (at a transcriptional level) to ex vivo stimulation. Meanwhile, GZMK+ cells quickly up-regulated an effector profile.

Leveraging scTCRseq we could track TCR clones before and after stimulation to understand their transcriptional response. We found that these Interferon-stimulated gene (ISG) cells were clonally related to a subset of CD8+ cells characterised by high GZMK expression.
