#HRH1

the mechanism from the paper is basically that COVID will bind to ACE2 and HRH1, using HRH1 to better bind to ACE2. antihistamines saturate HRH1 so that COVID can’t bind to it as easily, making infection and replication more difficult.

ACE2 and HRH1 (histamine 1 receptor) actually bind to each other, which can help COVID enter cells. So there's sort of 2 paths to entry, either through ACE2, or if there's not a lot of ACE2 receptors, through HRH1.

Antihistamines that block H1 could prevent COVID from entering cells!!

Antihistamines should be part of a defense in depth strategy and not your only precaution. Did I mention you should be masking? Because you should be masking! That having been said: Because the virus uses the HRH1 receptors to enter cells, keeping those receptors busy reduces the attack vector.

Research summary thread!!
A COVID research paper found that the histamine 1 receptor (HRH1) is highly involved in enhancing COVID infection.

They also found that most H1 blockers (antihistamines, like Claritin) reduced COVID after exposure!

One of the papers I need to add to the entry is journals.asm.org/doi/10.1128/... -- it needs an "in mice" caveat, but researchers have shown that SARS-CoV-2 actually does use the HRH1 pathway for infection in addition to hACE-2, and antihistamines keep HRH1 too busy for the virus to take hold.

This is a really big deal for a few reasons:
Initially, it was thought that the HRH1 (histamine 1 receptor) only enhanced the virus through working with the ACE2 receptor.

While it *can* do that, it can also enhance COVID without the ACE2 receptor at all!

"our results confirmed that HRH1 is directly bound to SARS-CoV-2 S and hACE2." They also tested various strains of COVID (including delta and BA variants) and found that all of them were affected by antihistamines that blocked the H1 receptor!

They focused on 5 antihistamines.

I want this so much. I've been waiting for next gen COVID vaccines for years, and I'm really hoping that this works better than the current ones, at least in terms of transmission and waning immunity over time. I'm also awaiting more research on the HRH1 route beyond that one paper from last year.

More evidence that HRH1 antagonist antihistamines can help prevent COVID.

There is an ever-increasing body of scientific evidence that the virus uses cells' histamine receptors for infection and that's part of why it's so virulent: journals.asm.org/doi/10.1128/... Large enough quantities of antihistamines serve as preferential binders and reduce capacity for viral entry.

It's looking like a combo of mast cell involvement and SARS-CoV-2 using the HRH1 receptors as alternate viral entry! I started telling even my non-MCAS friends who have terrible vaccine reactions to try the MCAS protocol of 50mg diphenhydramine 15m pre-vaccine --
April 30, 2025 at 8:00 PM
Yup, the spike protein is an extra potent mast cell destabilizer to the point it can cause MCAS reactions in people without MCAS! About 60% of long COVID cases improve significantly if you treat them like MCAS, and the virus uses the HRH1 receptor to enter cells and reproduce--

Add that paper about HRH1 as a potential secondary mechanism for infection to the implication that a lot of COVID-related symptoms are mast cell related and I am strongly on team "antihistamines are a useful COVID prevention measure" forevermore.
