Blocking IL1RAP may weaken pancreatic cancer’s treatment defenses by disrupting the inflammatory network around tumors. Preclinical results also showed less fibrosis and a better response to combination therapy. doi.org/hb8jpv
Blocking IL1RAP may weaken pancreatic cancer defenses before surgery
Blocking IL1RAP, a receptor that sits at a key control point in inflammatory signaling, can disrupt the tumor-driven inflammatory network that helps pancreatic cancer resist treatment, according to new research.

Alternatively spliced mini-exon B in PTPδ regulates excitatory synapses through cell-type-specific trans-synaptic PTPδ-IL1RAP interaction | Nature Communications
PTPδ, encoded by PTPRD, is implicated in various neurological, psychiatric, and neurodevelopmental disorders, but the underlying mechanisms remain unclear. PTPδ trans-synaptically interacts with multiple postsynaptic adhesion molecules, which involves its extracellular alternatively spliced mini-exons, meA and meB. While PTPδ-meA functions have been studied in vivo, PTPδ-meB has not been studied. Here, we report that, unlike homozygous PTPδ-meA-mutant mice, homozygous PTPδ-meB-mutant (Ptprd-meB–/–) mice show markedly reduced early postnatal survival. Heterozygous Ptprd-meB+/– male mice show behavioral abnormalities and decreased excitatory synaptic density and transmission in dentate gyrus granule cells (DG-GCs). Proteomic analyses identify decreased postsynaptic density levels of IL1RAP, a known trans-synaptic partner of meB-containing PTPδ. Accordingly, IL1RAP-mutant mice show decreased excitatory synaptic transmission in DG-GCs. Ptprd-meB+/– DG interneurons with minimal IL1RAP expre

Analysis of 9/11 first responders links toxic dust exposure to specific blood cell mutations, increasing leukemia risk and highlighting IL1RAP as a potential therapeutic target.
9/11 study shows how toxic exposures may lead to blood cancers
A study led by researchers at the National Cancer Institute-designated Montefiore Einstein Comprehensive Cancer Center (MECCC) has found that mutations in blood-forming cells may explain the increased risk for leukemia and other blood disorders among first responders exposed to the 9/11 World Trade Center (WTC) disaster site and its toxic dust.
