God I hate protein naming conventions.

“What’s this one called?”

“Smoothened.”

“SMOOTHENED?!”

“Yep.”

“What about that one?”

“MAP kinase.”

“Did you say MAP kinase?”

“Perhaps you’re thinking of MAP kinase kinase?”

“What??”

“Or MAP kinase kinase kinase?”

(Yes all of these are real proteins)

20 years ago my group published a paper that identified a target (a kinase) in pancreatic cancer that drove cell proliferation and chemotherapy resistance. We worked with a medicinal chemist who designed a small molecule inhibitor (SMI) to this kinase. In 2018, this SMI went into the clinic. 1/2

Now out in Science! Cyclin-dependent kinases (CDKs) are key regulators of the cell cycle. In @vcushing.bsky.social's magnum opus, we use #cryoEM to figure out how the CDK-activating kinase recognises CDKs to fully activate them - a key step in cell cycle control.

so the initial creatine kinase level may often be unremarkable.

Can we program a kinase like a switch? Inspired by natural autoinhibitory complexes, we designed miniproteins against active- and inactive-like conformations of Focal Adhesion Kinase. Depending on the targeted state, the resulting binders either activated or inhibited the kinase.

🚨 Our 4th example of metalloallosteric regulation of kinase activity. Out today in @CellReports with @KJSsci. The ER stress sensor kinase PERK needs copper to work. Without copper, PERK can’t protect cells under stress. cell.com/cell-reports/p…
