Reports of aggressive “giant ticks” in southern France prompted a 3-year survey (2,940 ticks).
The species was Hyalomma lusitanicum, carrying Rickettsia sibirica mongolitimonae. MALDI-TOF MS proved highly reliable for tick identification. 🧪

Enhancing Ion Yields in MALDI-2-MSI Using Dopant-Assisted Post Ionization for High-Resolution MS-Imaging of Lipids and Metabolites
AbstractMatrix-assisted laser desorption ionization mass spectrometry imaging (MALDI-MSI) is the most widely used label-free method for mapping the molecular distribution of lipids and metabolites from tissue sections and cell cultures. To mitigate the problem of partially low MALDI ion yields for several of these analyte classes, different post ionization techniques have recently been developed─for example, MALDI-2, where a second UV-laser pulse intersects the initial particle plume to generate additional charges. Here, we show that the MALDI-2 signal intensities of important analyte classes can be further increased upon introducing reactive dopant vapor into the MALDI-2 ion source. Dopant assistance as an optional add-on was realized in prototype form on MALDI-2 ion sources of a Spectroglyph Injector-Orbitrap MSI setup and a timsTOF fleX MALDI-2 instrument. Acetone, anisole, toluene, 2-propanol, and chlorobenzene were tested as dopants. Acetone proved to be the most effective and robust dopant for the given setup and MALDI matrix, and thus was used for further parameter optimization on both instruments. We demonstrate that with dopant-assisted (DA-)MALDI-2, a signal increase of up to 5 orders of magnitude, as compared to regular MALDI, is, for instance, achieved for several classes of sterols, glycerophospholipids, and sphingolipids. Next to the increased intensity of signals for abundant lipids, the total amount of annotated signals (based on MS1) was also majorly increased by the use of dopants. We demonstrate the potential for biomedical imaging applications with the examples of murine spleen and cerebellum sections, analyzed in the positive- and negative-ion mode.

Analysis of Carbohydrates and Glycoconjugates by Matrix‐Assisted Laser Desorption/Ionization Mass Spectrometry: An Update for 2023–2024
ABSTRACT The use of MALDI mass spectrometry for the analysis of carbohydrates and glycoconjugates is a well-established technique and this comprehensive review is the twelfth update of the original article published in 1999 and brings coverage of the literature to the end of 2024. Although still popular for glycoconjugate analysis, recent years have seen a slight drop in its usage with electrospray and LC-MS techniques becoming more widely used. The review follows the same general format as previous reviews. Much of the material relating to applications is presented in tabular form whereas papers relating to method development are discussed in more detail. The review is divided into three sections: (1) general aspects such as theory of the MALDI process, matrices, derivatization, MALDI imaging, fragmentation, quantification, and the use of computer software for structural identification. (2) Applications to various structural types such as oligo- and polysaccharides, glycoproteins, glycated proteins, glycolipids and natural products, and (3) other general areas such as medicine, biopharmaceuticals, industrial processes and glycan synthesis where MALDI is extensively used. As with previous reviews in this series, this review also includes a few papers that describe methods appropriate to analysis by MALDI, such as sample preparation, even though the ionization method is not MALDI. Much of this work is tabulated. MALDI is still an ideal technique for carbohydrate analysis, particularly in its rapidity, ease of use and its ability to produce single ions from each analyte. Improvements in the technique and range of applications show little sign of diminishing.
