In my experience [RNA] is pretty good on Nanodrop, it is [DNA] where it fails massively. We use Qubit for concentrations but Nanodrop can be useful for 260/280 and 260/230

At first I was like “I’m not gonna pay 5 more dollars for plasmid extraction and sequencing in less than 24 hours”….but…it’s so much easier and it magically works way better than having undergrads prep and nanodrop
I am thankful for gene synthesis, hi-fi assembly, and inexpensive whole-plasmid sequencing.

Forse non tutti sanno che... In laboratorio, in inverno, giro con un'asticella di metallo in tasca per scaricare l'elettricità statica perché, a causa dei miei vestiti in poliestere da povero, prendo continuamente la scossa e una volta ho fatto saltare il PC del Nanodrop 🥲

The story of NanoDrop is so interesting. When it first came on the scene, it was very good at what it did. Then people started using it outside that range, slowly realizing it is then a random number generator.

Starting with a regular Q5 run without additives. 30ng/uL template gDNA from a proprietary actinobacteria I'm developing for @SenseoryPlants. Dimers and faint bands, despite clear sign of gDNA presence in wells and nanodrop looks workable. These cells are hard to crack!

Took my kid to a science festival at the weekend, she was absolutely over the moon to do a ‘real experiment’ extracting DNA (from some blueberries), just like her dad.
She’s going to be absolutely devastated when I sit her down and explain how rubbish her absorbance ratios are.

I was always told that the nanodrop can’t accurately measure gDNA, but in my hands it absolutely can, if it’s cleanly isolated and you take 30 seconds to needle shear your sample first.

We've measured bacterial RNA with nanodrop, qbit, and bioanalyzer.
All three give distinct values and none are well correlated.
What's the generally accepted standard for measuring RNA concentrations?
