T and B cells recognize splicing-derived neutrophil neoantigens in lupus - Nature
In systemic lupus erythematosus, aberrant splicing in neutrophils generates novel peptides that are presented on MHC I, driving CD4+ and CD8+ T cell activation, clonal expansion and IgG autoantibody responses, establishing neoantigen-driven autoimmune immunity.

Removing neutrophils reduced lung metastases in mice. The decline came with fewer “hybrid” cancer cells—the intermediate state most likely to spread. doi.org/hcq385
Bad neighbors: How immune cells help cancer cells get ready to spread
Most cancer deaths are not caused by the original tumor, but by metastases, which form when cancer cells leave the original tumor and spread to other organs.

Correlation isn’t enough, so we tested it.

Deplete neutrophils or block OSM → the hybrid state collapses, cells shift toward late EMT, and lung metastasis drops.

The niche doesn’t just mark the state, it actively maintains it.

Why? The neighbors talk back.

Neutrophils secrete oncostatin M (OSM). OSM does something clever: it switches on mesenchymal genes (SNAI1, ZEB1) while keeping the epithelial program (∆Np63, KLF5) alive.

The result: cells held in a hybrid state.

This paper proposes a physical mechanism by which fragments of SARS-CoV-2 spike, cut by host proteases, can selectively kill microvilli-rich immune cells (pDCs, T cells) while sparing smooth monocytes and neutrophils. It is a mechanistic in vitro study of peptides and healthy-donor cells.

And the niches are self-reinforcing. 🔁

OSM makes tumor cells pump out neutrophil-recruiting signals → more neutrophils → more OSM.
Late-EMT cells recruit more macrophages → more TGFβ1.

Each signal builds the very niche that sustains it.

Jiang, Bapat &co show @nature.com that eating a fat diet in proximity (4 days!) of a skin inflammatory insult exacerbates Th17 responses in lean mice by recruiting neutrophils that produce IL1b via NLRP3, also epigenetically reprogramming Trm hyper responsive cells!

5) Neutrophils can also promote clotting by expelling DNA and proteins in web called Neutrophil extracellular traps (NETs). Here they tested NETosis measures such as Myeloperoxidase and Citrullinated H3.

All of these measures showed NO significant difference with controls.

Dengue Virus Serotype 2 Infection Transcriptionally Reprograms Host Neutrophils to Establish an Immunosuppressive Microenvironment in Mice bioRxivpreprint
Dengue Virus Serotype 2 Infection Transcriptionally Reprograms Host Neutrophils to Establish an Immunosuppressive Microenvironment in Mice
Dengue virus (DV) infection triggers profound alterations in host immune responses, yet the transcriptomic reprogramming of innate immune cells during disease progression remains incompletely understood. Here, we demonstrated that Dengue virus serotype 2 (DV-2) infection transcriptionally reprograms host neutrophils, establishing an immunosuppressive microenvironment that facilitates pathogenesis. Through scRNA transcriptomic profiling of bone marrow and spleen cells from the DV-2-infected AG129 mouse model, we delineated a distinct neutrophil signature that shows dynamic shifts in immature and activated populations over the course of dengue infection. We also characterized a specific Ly6Glow subpopulation, notably Fibrinogen-like protein 2 (FGL2)-expressing subsets, that appeared with disease progression. Serum FGL2 levels were significantly elevated in both DV-2-infected mice and human dengue-positive patients. Furthermore, treatment of splenocytes with FGL2 or FGL2-enriched neutrophil secretomes suppressed T cell functional activity and promoted an immunosuppressive phenotype. Together, these findings reveal how DV-2-driven neutrophil reprogramming shapes an immunosuppressive network, highlighting the importance of studying FGL2-expressing neutrophil subsets to understand dengue immunopathogenesis.
