We differentiated mESCs into mature forebrain organoids. They express excitatory and inhibitory synapse markers (Vglut1, Vgat, Psd95, Gephyrin) and show spontaneous calcium activity that TTX blocks, indicating action-potential-dependent network activity. 2/5

In mouse hippocampal CA1, synapse-related markers followed different developmental and prenatal-valproate patterns depending on layer and dorsoventral position.

And we can keep adding layers, like labelling proteins (e.g. PSD95 in PRISM and LICONN). I think the endgame is that the connectome data will be so rich that it'll be conceptually a different kind of object, an ultrastructome. 3/

We find that short-lifetime PSD95 synapses (but not long-lifetime PSD95 nor SAP102) increase sc-fc coupling _specifically_ in awake (not anaesthetized) mice.

To better understand the mechanisms behind this process we teamed up with @cianodonnell.bsky.social and a great student in his lab @jajmca.bsky.social who helped us model the incorporation of PSD95 into nascent synapses and how this influences their number, strength and stability.
