Why do some HEK293 cells crank out more viral vectors than others? 🧬

We used multi-omics + metabolic modeling to find out. Turns out, pseudohypoxia is a big deal.

So we built condition-specific genome-scale metabolic models and integrated lipidomics, exometabolomics, and transcriptomics

What stood out?

HIF-1α was active in the low-producer—driving that pseudohypoxia

So we tried inhibiting HIF-1α and ...

capsid yield jumped 2.5x 🎉

But... there’s a catch.

More mad thoughts: In naked mole rats, a tuber-based diet and protected subterranean life may interact with hypoxic signalling. In humans, increased starch consumption and better shelter/safety occurred without chronic hypoxia, but phytochemicals may have provided mild pseudohypoxia-like signals.

Oocytes develop in hypoxia with active HIF signaling, which is linked to hyaluronan pathways. More actual hypoxia probably isn’t helpful, but plant defence metabolites can affect HIF and cause pseudohypoxia. Could that increase HAS2 expression and protective HMW-HA and set it like that?

Could our neoteny be historically linked to diet?
Some plant defence metabolites can induce pseudohypoxia, while others affect hypoxia-related signalling pathways.

This doesn’t require infection.

Physiological states that raise HA turnover include:
• elevated insulin
• elevated estrogen
• hypoxia / pseudohypoxia (e.g. preeclampsia)

All increase the chance of HA fragmentation.
