Loss of GPC6 generates a molecular hybrid of 2 developmental diseases: WNT5A deficiency characteristic of Robinow syndrome and pathological ERK activation characteristic of the RASopathies. Omodysplasia caused by loss of GPC6 overlaps both disease classes. #omodysplasia #RASopathy #Robinow

Interesting! So in RASopathies conditions, genetically elevated RAS/MAPK signaling could theoretically prevent or weaken DNMT remodeling in response to actin disruption/wounding. Predict poor wound healing? Clinically, ppl with rasopathy have altered ectodermal and keratinocyte manifestations.

🧪Excited to dig into this paper. One outstanding question in the #RASopathies field is how different variants in SHP2 cause two distinct conditions #Noonansyndrome (NS) and Noonan syndrome with Multiple Lentigines (NSML) AND how they respond to drug treatments (mTORi vs MEKi)
New research from the @jlmcfalinefigueroa.bsky.social and @nshahlab.bsky.social groups, recently published in Science Advances, uncovers novel mechanisms by which SHP2 mutations disrupt cellular processes. This work may help pave the way toward future therapies for patients > bit.ly/4nGsVZm
