Not true - zuranolone (PPD), suzetrigine (pain), gepants (migraine), and more... were born out of a long arc of studies in the lab.

On Thursday, the US Food and Drug Administration approved suzetrigine for short-term pain management, making it the first non-opioid pain drug given a regulatory nod in more than 20 years.

Unlike opioids, which work by latching onto receptor proteins on nerves in the central nervous system, suzetrigine works only in peripheral nerves—that is, those outside the brain and spinal cord.

The good news is there's a new option for acute/postsurgical pain (suzetrigine) that is not an opioid! The bad news is it doesn't work on chronic pain :/

Suzetrigine is a sodium channel inhibitor that suppresses the ability of nerves to communicate pain signals to the brain. (It also says really interesting things about the difference between acute and chronic pain for it only to work on one of them.)

I would honestly recommend poor metabolizers push their insurance to cover suzetrigine, which is only useful in acute/post-surgical pain, not chronic, but is a different metabolism and also not an opiate.

Exciting new work on #suzetrigine effects on spontaneously active human DRG neurons from patients/donors with a pain history. Short story is that suzetrigine is remarkably effective at blocking spontaneous activity. Collab with Dougherty lab at MDACC!

In clinical studies (pubmed.ncbi.nlm.nih.gov/37530822/), Suzetrigine reduced pain more effectively than placebo in patients undergoing “tummy tuck” or bunion surgery.

Also in the mix: suzetrigine, a new non-opioid painkiller. It blocks a sodium channel (NaV1.8) on peripheral pain nerves, so signals never reach the brain. First genuinely new class of analgesic in over 25 years, after decades of targeting the same pathway.

from what I understand the main usage case for suzetrigine/ journavx is surgery recovery, so if you have that coming up and want to avoid opioids that might be a good thing for you. expensive as fuck though

Never forget about training. The 1st author on the suzetrigine paper got his start in research as a postbac in the intramural program at NIH, a PhD at Columbia and a postdoc with Nobel laureate (although not at the time) David Julius at UCSF.
