Combatting my prejudices starts with identifying and disclosing my biases.

I apologize to those - past, present, & future - who I have judged harshly bc they wrongly referred to TNF as “TNFalpha”.

Some of them are serious immunologists … probably… I guess.

persönlich halte ich GSK3 Inhibition für einen wichtigen Ansatzpunkt. Nicht zwingend den einzigen. Evtl in Kombination mit Low Dose mTOR und oder TNFalpha Inhibitoren.

Viele Nahrungsmittel eigen sich zum testen, so enthält Pfeffer zum Beispiel einen CB2 Agonisten, Holundersaft eignet sich als mTOR Inhibitor, Lithium ist ein GSK3 Inhibitor, Kaffeinsäure inhibiert vermutlich TNFalpha, Kapern enthalten den MastZell-Stabilisator Quercetin.... Da lässt sich

absolutely incredible: complete TNFalpha deficiency (via homozygous frameshift) causes recurrent pulmonary TB (via impaired NADPH oxidase activity in macrophages) but not much else: www.nature.com/articles/s41...

it's orders of magnitude milder of a phenotype than i'dve expected.

Yes. If the infection is too far ahead of the immune system, you can get uncontrolled activation leading to massive cytokine release that also stimulates the release of cytokines more systemically. The liver is a huge source of inflammatory cytokines - IL-1beta, TNFalpha and IL-6.

habt ihr es schon mit höherer Ballaststoff Zufuhr vor+während der Depressionsphase versucht (z.b. täglich 1/4 Knolle Fenchel o.ä.)?

und darüber hinaus, TNFalpha Level sind meines Wissens Zyklus abhängig und wirken auf IDO 🤔
vor allem kann man mit diesem Wissen wenigstens ein bisschen Gegensteuern. Im Fall von Kynurein Stoffwechsel z.b. mit Ballaststoffen. Die Bakterien in unserem Darm produzieren daraus Butyrat, welches IDO (1.Schritt Kynurein Stoffwechsel) runter reguliert. Einige Ballaststoff-Beispiele:

If I had to guess, I'd say it's a cytokine that also fluctuates in response to hormonal changes, such as TNFalpha.

In addition to wrongly claiming T cell exhaustion (wrong, T cells are fit & make cytokines), reduced CD4 T cells & increased CD8 T cells making TNFalpha (not what data shows), your team is now engaging with pseudoscience. It gives false hope and erodes trust in science bsky.app/profile/marc...

Roughly 20% to 30% of people with Crohn's will not respond to the other more well established medications. This puts us at 20K to 30K people, nationwide needing this specific medication, or similar (non-TNFalpha blocker immunosuppressant; They're priced similarly to Entyvio).
