**Abstract:** This research introduces a novel approach to dynamically characterize the nanometer-scale reorganization of connexin43 (Cx43) tetrameric complexes within gap junctions, a crucial process governing intercellular communication. Leveraging advanced volumetric deep learning reconstruction techniques applied to super-resolution microscopy data, we develop a robust methodology capable of identifying and tracking conformational changes in Cx43 tetramers with unprecedented spatial and temporal resolution. This framework provides a quantitative tool for understanding the biophysical mechanisms regulating gap junction permeability and highlights potential therapeutic targets for diseases characterized by aberrant intercellular coupling.
